The short version

Skin takes on colour in two very different ways, and the compounds on this desk sit on opposite sides of that split.

The first route is chemical and local. Copper peptides — the best studied is GHK-Cu, a three-amino-acid molecule holding a single copper ion — are worked into creams and serums. Copper is itself a coloured thing, and it is a helper metal (a cofactor) for the enzyme that manufactures pigment. Under the ingredient name Copper Tripeptide-1, GHK-Cu sits legally on cosmetic shelves in the United States, the United Kingdom and the European Union.

The second route is hormonal and whole-body. Melanotan II is an injected copy of a hormone that tells pigment cells to switch on, so skin darkens without sunlight. It has never been approved anywhere for anything. The published record around it includes moles that darken and multiply, kidney injury, and prolonged painful erections.

Same shelf in marketing. Different worlds in pharmacology, and different worlds in law.

Two routes to colour

The copper route is a matter of chemistry sitting in the skin. GHK-Cu is the copper(II) chelate of glycyl-L-histidyl-L-lysine, a sequence that occurs naturally inside the alpha-2(I) chain of type I collagen. The copper ion is not decoration: it is what allows lysyl oxidase to cross-link collagen and elastin, and it gives the complex a superoxide-dismutase-like antioxidant character. In fibroblast culture the peptide raises collagen synthesis without raising cell number, an effect that begins at picomolar concentrations and peaks around a nanomolar one [7]. Its acknowledged problem is delivery — the free peptide is strongly water-loving (clogP -2.24), which is exactly the wrong property for crossing the stratum corneum [1].

The melanocortin route is a matter of signalling reaching the whole organism. Melanotan II is a cyclic, protease-resistant analogue of alpha-melanocyte-stimulating hormone. It binds MC1R on the melanocyte, raises cyclic AMP, and drives the PKA-CREB-MITF cascade that upregulates tyrosinase and pushes pigment synthesis toward eumelanin — colour without ultraviolet light. But the same molecule is non-selective across MC1R through MC5R, so the appetite, sexual and cardiovascular receptors are engaged at the same time. In a double-blind placebo-controlled crossover study in ten men, a subcutaneous dose produced clinically apparent erections in eight of them [12]. A tanning compound is never only a tanning compound.

The frame this site keeps returning to is that a shared word — peptide, or glow — is doing an enormous amount of quiet work across that gap.

Where the regulatory line actually falls

It is tempting to read a cosmetic ingredient list as a kind of safety certificate, and to read the absence of a drug approval as a verdict of danger. Both readings are wrong, and the error runs in both directions.

An INCI name such as Copper Tripeptide-1 is a naming convention. Finding it on a serum means the ingredient is permitted in cosmetics in that jurisdiction and that the product is sold on appearance claims. It does not mean the molecule has been evaluated as a medicine, and it says nothing whatsoever about what happens when the same molecule is reconstituted and injected. GHK-Cu has no approved therapeutic indication by any route, in any jurisdiction; systemic use sits outside the cosmetic record entirely and has no validated human pharmacokinetics behind it.

Running the other way: Melanotan II's total absence of approval is not softened by the approvals sitting next to it. Afamelanotide, the linear analogue, is separately approved for a rare light-sensitivity disorder, and bremelanotide, derived from Melanotan II itself, is approved for a sexual-desire indication [11]. Those approvals belong to those molecules and their trial packages. Family resemblance launders nothing.

Regulatory status attaches to a route and a claim, not to a molecule's reputation.

What research peptide means on this desk

Peptides are short chains of amino acids — the same alphabet proteins are written in, just far fewer letters. That brevity is why they are attractive to formulators and to researchers: a short sequence can be synthesised cleanly, and a well-chosen one can imitate a signal the body already uses.

It is also why the category is so slippery. A tripeptide in a cosmetic emulsion, a heptapeptide analogue of a pituitary hormone, and a supplier-mixed injectable blend all answer to the word peptide while sharing almost nothing else — not the evidence base, not the route, not the regulatory register, not the risk.

A 2026 review of approved and unapproved peptide therapies puts the position plainly: many of these compounds show favourable tissue-repair outcomes in animal models while rigorous human safety data stay scarce, in a grey market operating largely outside regulatory oversight [13]. That sentence is the honest frame for everything here.
